PROTEIN SUMMARY GENE INFORMATION RNA DATA ANTIBODY DATA
GENERAL INFORMATIONi

General description of the gene and the encoded protein(s) using information from HGNC and Ensembl, as well as predictions made by the Human Protein Atlas project.

Gene namei

Official gene symbol, which is typically a short form of the gene name, according to HGNC.

CTCF
Synonyms CFAP108, FAP108
Gene descriptioni

Full gene name according to HGNC.

CCCTC-binding factor
Protein classi

Assigned HPA protein class(es) for the encoded protein(s).

Cancer-related genes
Disease related genes
Human disease related genes
Transcription factors
Predicted locationi

All transcripts of all genes have been analyzed regarding the location(s) of corresponding protein based on prediction methods for signal peptides and transmembrane regions.

  • Genes with at least one transcript predicted to encode a secreted protein, according to prediction methods or to UniProt location data, have been further annotated and classified with the aim to determine if the corresponding protein(s) are secreted or actually retained in intracellular locations or membrane-attached.

  • Remaining genes, with no transcript predicted to encode a secreted protein, will be assigned the prediction-based location(s).

The annotated location overrules the predicted location, so that a gene encoding a predicted secreted protein that has been annotated as intracellular will have intracellular as the final location.

PROTEIN FUNCTION
Protein function (UniProt)i

Useful information about the protein provided by UniProt.

Chromatin binding factor that binds to DNA sequence specific sites and regulates the 3D structure of chromatin 1, 2, 3. Binds together strands of DNA, thus forming chromatin loops, and anchors DNA to cellular structures, such as the nuclear lamina 4, 5, 6. Defines the boundaries between active and heterochromatic DNA via binding to chromatin insulators, thereby preventing interaction between promoter and nearby enhancers and silencers 7, 8, 9. Plays a critical role in the epigenetic regulation 10. Participates in the allele-specific gene expression at the imprinted IGF2/H19 gene locus 11, 12, 13. On the maternal allele, binding within the H19 imprinting control region (ICR) mediates maternally inherited higher-order chromatin conformation to restrict enhancer access to IGF2 (By similarity). Mediates interchromosomal association between IGF2/H19 and WSB1/NF1 and may direct distant DNA segments to a common transcription factory (By similarity). Regulates asynchronous replication of IGF2/H19 (By similarity). Plays a critical role in gene silencing over considerable distances in the genome (By similarity). Preferentially interacts with unmethylated DNA, preventing spreading of CpG methylation and maintaining methylation-free zones 14. Inversely, binding to target sites is prevented by CpG methylation 15. Plays an important role in chromatin remodeling 16. Can dimerize when it is bound to different DNA sequences, mediating long-range chromatin looping 17. Causes local loss of histone acetylation and gain of histone methylation in the beta-globin locus, without affecting transcription 18. When bound to chromatin, it provides an anchor point for nucleosomes positioning 19. Seems to be essential for homologous X-chromosome pairing (By similarity). May participate with Tsix in establishing a regulatable epigenetic switch for X chromosome inactivation 20. May play a role in preventing the propagation of stable methylation at the escape genes from X-inactivation 21. Involved in sister chromatid cohesion 22. Associates with both centromeres and chromosomal arms during metaphase and required for cohesin localization to CTCF sites 23. Plays a role in the recruitment of CENPE to the pericentromeric/centromeric regions of the chromosome during mitosis 24. Acts as a transcriptional repressor binding to promoters of vertebrate MYC gene and BAG1 gene 25, 26, 27. Also binds to the PLK and PIM1 promoters 28. Acts as a transcriptional activator of APP 29. Regulates APOA1/C3/A4/A5 gene cluster and controls MHC class II gene expression 30, 31. Plays an essential role in oocyte and preimplantation embryo development by activating or repressing transcription (By similarity). Seems to act as tumor suppressor 32.... show less

Gene summary (Entrez)i

Useful information about the gene from Entrez

This gene is a member of the BORIS + CTCF gene family and encodes a transcriptional regulator protein with 11 highly conserved zinc finger (ZF) domains. This nuclear protein is able to use different combinations of the ZF domains to bind different DNA target sequences and proteins. Depending upon the context of the site, the protein can bind a histone acetyltransferase (HAT)-containing complex and function as a transcriptional activator or bind a histone deacetylase (HDAC)-containing complex and function as a transcriptional repressor. If the protein is bound to a transcriptional insulator element, it can block communication between enhancers and upstream promoters, thereby regulating imprinted expression. Mutations in this gene have been associated with invasive breast cancers, prostate cancers, and Wilms' tumors. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2010]... show less
PROTEIN INFORMATIONi

The protein information section displays alternative protein-coding transcripts (splice variants) encoded by this gene according to the Ensembl database.

The Splice variant identifier links to the Ensembl website protein summary for the selected splice variant. The data in the Swissprot and TrEMBL columns links to corresponding pages in the UniProt database.

The protein classes assigned to this protein are shown if expanding the data in the protein class column. Parent protein classes are in bold font and subclasses are listed under the parent class.

The length of the protein (amino acid residues according to Ensembl), molecular mass (kDalton), predicted signal peptide and number of predicted transmembrane region(s) according to in-house majority decision methods based on sets of predictors are also reported.
